稳斑汤对ApoE基因敲除小鼠主动脉粥样硬化不稳定斑块形成Bax和Bcl-2 mRNA的影响 被引量:8

摘要 目的观察搜风祛痰法中药复方稳斑汤对Apo E基因敲除小鼠主动脉粥样硬化不稳定斑块形成Bax和Bcl-2 mRNA的影响。方法选取6~8周Apo E基因敲除小鼠建立动脉粥样硬化(AS)不稳定斑块模型,随机分为模型组,西药组,稳斑汤低、中、高剂量组,另取正常小鼠做空白对照组。空白对照组和模型组分别给予生理盐水灌胃,西药组及稳斑汤低、中、高剂量组分别给予阿托伐他汀及不同剂量稳斑汤干预。取小鼠主动脉组织HE染色进行病理学观察,并采用半定量RT-PCR技术检测Bax和Bcl-2 mRNA表达的变化。结果与空白组比较,模型组Bax mRNA表达水平明显增高(P<0.05),Bcl-2 mRNA表达水平明显降低(P<0.05);与模型组比较,稳斑汤各剂量组及西药组Bax mRNA表达水平明显降低(P<0.05),Bcl-2 mRNA表达水平明显增高(P<0.05);稳斑汤高剂量组与西药组比较Bax mRNA、Bcl-2 mRNA表达水平无差异(P>0.05)。光镜下观察稳斑汤各剂量组和西药组斑块面积均小于模型组。结论搜风祛痰法复方中药稳斑汤治疗AS的机制可能与抑制Bax mRNA的表达,促进Bcl-2 mRNA表达有关。 Objective To observe the effect of compound plaque- stablizing decoction on unstable plaque Bax mRNA and Bcl- 2 mRNA expression of Apo E knockout rats with Atherosclerosis. Methods A number of 6 to 8 weeks old Apo E gene- deficient rats were selected to establish AS unstable plaque model,and were randomly divided into the model group,the western medicine statin group and the stable plaque decoction low,medium and high dose groups,and the control group. They were given normal saline,low dose,medium dose,high dose compound stabilizing plaque decoction,and Atorvastatin for a month. Aortic tissue was observed under optical microscope pathology after HE staining. Detect the expression levels of Bax mRNA and Bcl- 2 mRNA for each group with RT- PCR. Results Bax mRNA expression levels in the model group was significantly higher than the control group( P < 0. 05); Compared with model group,Bax mRNA expression levels in western medicine statin group and all stabilizing plaque decoction groups were significantly lower( P < 0. 05). There was no difference between western medicine statin group and stable plaque decoction high dose group in expression levels of Bax mRNA( P > 0. 05). Compared with western medicine statin group,Bax mRNA expression levels in the stable plaque decoction low and medium dose groups were significantly higher( P < 0. 05). Bcl- 2 mRNA expression levels in the model group was significantly lower than the control group( P < 0. 05); Compared with model group,Bax mRNA expression levels in western medicine statin group and all stabilizing plaque decoction groups were significantly higher( P < 0. 05). There was no difference between western medicine statin group and stable plaque decoction high dose group in expression levels of Bax mRNA( P > 0. 05). Compared with western medicine statin group,Bax mRNA expression levels in the stable plaque decoction low and medium dose groups were significantly higher( P < 0. 05). The plaque area in each dose group and western medicine group were significantly smaller than that of the model group by light microscope. Conclusion Searching wind and eliminating phlegm compound plaque- stabilizing decoction may stabilize plaques by restraining Bax expression and promoting Bcl- 2 expression,and its mechanism may be associated with its protection for the cardiovascular system.
