火针对带状疱疹后遗神经痛大鼠疼痛阈值及NF-κB表达的影响 被引量:13

The Effects of Ignipuncture on Pain Threshold and NF-κB Expression in Rats with Postherpetic Neuralgia

摘要 目的观察火针对带状疱疹后遗神经痛大鼠机械性痛阈值、脊髓核转录因子-κB(nuclear factor-κB,NF-κB)表达的影响,探讨火针治疗带状疱疹后遗神经痛的可能机制。方法将30只雄性SD大鼠,随机分为3组,即空白对照组、模型组、火针组,每组大鼠均为10只。通过VZV接种建立带状疱疹后神经痛(postherpetic neuralgia,PHN)动物模型。于接种前1 d,接种后1 d、4 d、7 d、14d和21 d分别进行机械性痛阈(paw withdrawal threshold,PWT)测定。接种后第7天开始,火针组进行火针治疗,隔日治疗1次,7次1个疗程。第21天,取各组大鼠L4、5段脊髓,用免疫组化法测定脊髓NF-κBp65蛋白表达情况。结果与空白对照组比较,模型组从造模后7 d开始出现PWT值下降(P<0.01),造模后第14天、21天时仍处于较低水平(P<0.01)。与模型组比较,火针组第7天治疗后开始出现PWT值上升(P<0.01),第14天、21天时PWT值上升明显(P<0.01)。与空白对照组比较,模型组脊髓组织中NF-κBp65表达明显增多,差异有统计学意义(P<0.01),与模型组比较,火针组脊髓组织中NF-κBp65表达明显减低,差异有统计学意义(P<0.01)。结论火针可缓解PHN大鼠PWT值,抑制NF-κB的表达,可能是火针治疗PHN的机制之一。 Objective It is to investigate the effects of ignipuncture on pain threshold and NF- κB expression in rats with postherpetic neuralgia and to discuss the possible mechanism of ignipuncture for treatment of postherpetic neuralgia. Methods Thirty male SD rats were randomly divided into control group( C),model group( M),and ignipuncture group,with 10 cases in each. This subject by means of establishing PHN animal models by VZV vaccinated. 1stday before VZV vaccination,1stday,4thday,7thday,14 thday and 21 st day after VZV vaccination,respectively,mechanical pain threshold( von Frey filaments method),the paw withdrawal threshold( PWT) were measured respectively. On 7thday,ignipucture acupuncture was applied to ignipucture group after vaccination modeling,the treatment was given every other day,7 times a course. The21 stday,the rats were taken L4- 5for spinal cord,spinal NF- κB p65 measured protein expression by immunohistochemistry. Results In comparison with control group,PWT decreased significantly from the 7th day in model group( P < 0. 01),after 14 thday,21stday is still at a low level( P < 0. 01). In compared with the model group,PWT increased significantly from the 7thday in model group( P < 0. 01),after 14 thday,21stday PWT values increased significantly( P < 0. 01). In compared with the control group,model group NF- κB p65 was significantly increased in spinal cord tissue( P < 0. 01),in compared with the model group,ignipucture group was significantly reduced NF- κB p65 in spinal cord tissue( P < 0. 01). Conclusion The ignipucture has a significant analgesic effect in PHN rats,which can relieve PHN rats PWT value,inhibit the expression of NF- κB of the spinal cord,and it may be one of the mechanisms of ignipucture treatment of PHN.
