益气通脉胶囊对脑缺血大鼠学习记忆能力的影响 被引量:3

摘要 目的观察益气通脉胶囊对脑缺血损伤后大鼠学习记忆能力的影响及相关机理研究。方法选择SD大鼠90只,按随机数字表法选择10只作为对照组,其余大鼠采用反复夹闭双侧颈总动脉结合硝普钠降压法复制SD大鼠拟血管性痴呆模型。造模后选存活大鼠50只随机分为模型组、脑心通组和益气通脉胶囊高、中、低剂量组,每组各10只,连续给药15 d后通过Morris水迷宫测试大鼠的学习记忆能力,之后取大鼠脑的海马组织,提取蛋白测定生长相关蛋白43(growth associated protein 43,GAP43)和神经丝蛋白-200(Neurofilaments Protein-200,NF200)的表达,以及测定大鼠海马组织NOS活性和NO含量。结果 (1)大鼠的学习记忆能力:益气通脉胶囊高、中剂量组大鼠的学习记忆能力均高于模型组。(2)GAP43和NF200的表达:益气通脉胶囊高、中、低剂量组的蛋白表达均明显大于模型组。(3)益气通脉胶囊高、中剂量组大鼠的NOS活性和NO含量均显著低于模型组。结论益气通脉胶囊对脑缺血损伤后大鼠学习记忆能力的恢复有明显的促进作用,能使神经可塑性相关蛋白表达上调,降低大鼠海马组织NOS活性和NO含量。 Objective To observe the effects of Yiqitongmai Capsule on learning and memory abilities of rats with cerebral ischemia and study the mechanism. Methods Totally 90 healthy SD rats were randomized into control group( n = 10) and vascular dementia models,which were duplicated by repeated clamping of bilateral common carotid artery combined with sodium nitroprusside depressing. Then models were divided into 5 groups: model group,Naoxintong group and experimental groups including high dose,medium dose and low dose with 10 rats in each. 15 days of continuous infusion later,Morris water maze was applied to test the learning and memory abilities. Then,extracting the total protein from rat hippocampus and detecting the expression of GAP43 and NF200,and determining the activity of NOS and the content of NO in hippocampus of rats. Results( 1) Learning and memory abilities of rat in the experimental groups was higher than in the model group.( 2) The expression level of GAP- 43 and NF- 200 of all the experimental groups was more than those of the model.( 3) The activity of NOS and the content of NO in the experimental of high dose and medium dose were lower than those of the model. Conclusions Yiqitongmai Capsule is obviously effective on improving the learning and memory abilities in rats with cerebral ischemia. Meanwhile,it would increase the expression of neuronal plasticity associated proteins in hippocampus,and decrease the activity of NOS and the content of NO.
