苦酸通调方对自发性糖尿病大鼠腹部脂肪组织中NF-κB及IKKβ蛋白表达的影响 被引量:3

摘要 目的观察苦酸通调方对自发性糖尿病大鼠腹部脂肪组织中NF-κB及IKKβ蛋白表达的影响。方法将高脂饲料喂养的雄性SPF级ZDF大鼠30只,随机分为模型组、苦酸通调组、吡格列酮组3组,普通饲料喂养的雄性SPF级ZL大鼠为正常对照组,每组各10只。苦酸通调方组按照3.29 g/(kg·d)灌胃,吡格列酮组按照1.07 mg/(kg·d)灌胃。模型组和正常对照组灌服等体积蒸馏水,每天1次,连续12周。经Western blot法检测各组大鼠腹部脂肪组织中NF-κB、IKKβ蛋白的表达水平。结果在治疗第12周末,模型组、吡格列酮组、苦酸通调组大鼠腹腔脂肪组织中NF-κB、IKKβ蛋白表达明显高于正常对照组(P<0.01)。与模型组相比,苦酸通调组和吡格列酮组NF-κB、IKKβ蛋白的表达均明显降低(P<0.01,P<0.05),且苦酸通调组优于吡格列酮组(P<0.05)。结论苦酸通调方组在一定程度上能抑制糖尿病大鼠腹部脂肪组织中NF-κB、IKKβ蛋白的表达,可能是苦酸通调方抑制炎症信号通路的传导而发挥其改善胰岛素抵抗治疗糖尿病的机制之一。 Objective To investigate the effect of Kusuantongtiao Formula on the expression of NF-κB and IKKβ protein in abdominal adipose tissue of spontaneously diabetic rats. Methods The thirty male Zucker rats( Zucker Diabetic Fatty rats) which were induced by feeding of high-fat and high-carbohydrate diet were randomly divided into the model group( MG),the Pioglitazone group( PG) and the Kusuantongtiao group( KG). A normal control group( NG) was also set up by male Zucker Lean( ZL) rats which were feeding of normal diet. There were 10 rats in each group. The Kusuantongtiao group and the Pioglitazone group were given Kusuantongtiao decoction 3. 29 g /( kg·d) and Pioglitazone suspension 1. 07 mg /( kg·d) gavage respectively,while the normal control group and model group were given the same amount of normal saline.After twelve weeks,the expression of NF-κB and IKKβ protein in abdominal adipose tissue of spontaneously diabetic rats were detected by Western blot method. Result At the end of the twelfth week,the expression of NF-κB and IKKβ protein in abdominal adipose tissue was significantly higher than that in normal control group( P < 0. 01). Compared with MG,levels of the expression of NF-κB and IKKβprotein were decreased obviously( P < 0. 01,P < 0. 05). And the group of KG is significantly better than the group of PG( P <0. 05). Conclusion Kusuantongtiao Formula can inhibit the expression of NF-κB and IKKβprotein in abdominal adipose tissue of diabetic rats. And it may be the possible mechanism of Kusuantongtiao Formula preventing and controlling type 2 diabetes.
