温肺通痹颗粒对肺成纤维细胞增殖及细胞周期的影响

摘要 目的通过离体细胞实验,以MRC-5人胚肺成纤维细胞为研究对象,检测温肺通痹颗粒对MRC-5细胞的增殖及细胞周期的影响。方法将温肺通痹颗粒配制成终浓度为0.031、0.125、0.5、2、8 mg/ml的溶液,用MTT法测定细胞存活率及细胞存活周期。结果温肺通痹颗粒与MRC-5人胚肺成纤维细胞共孵育对细胞的增殖具有抑制作用,且作用效果与药物浓度及共孵育时间具有显著相关性;温肺通痹颗粒与MRC-5人胚肺成纤维细胞共同孵育,随着药物浓度的增大和作用时间的延长,G1期细胞的比例逐渐升高,G2期与S期相应降低,与对照组比较差异具有统计学意义,且呈现明显量效关系。结论温肺通痹颗粒对MRC-5人胚肺成纤维细胞具有明显抑制增殖的活性;温肺通痹颗粒对细胞的毒性非常小;温肺通痹颗粒可能具有稳定肺成纤维细胞,并抑制其大量增殖的作用。 Objective To explore the impacts of wenfei tongbi granule on MRC- 5 proliferation and cell cycle by in vitro experiment,taking MRC- 5 human lung fibroblasts as the subjects. Methods Preparing wenfei tongbi granules as the solution of the final concentration of 0. 031,0. 125,0. 5,2 and 8 mg / ml.MTT method was used to determine the cell survival rate and the cell survival cycle. Results Incubated with wenfei tongbi granules,the cell proliferation was inhibited,presenting the significant correlation with the drug concentration and incubation time. The effect time was prolonged when the drug concentration was increased.The cell proportion at G1 phase was increased gradually and that at G2 phase and S phase was reduced,indicating the significant difference as compared with the control group. It displayed the apparent dose- effect relationship. Conclusion Wenfei tongbi granules obviously inhibit the activity of MRC- 5 proliferation,with very low toxicity. The granules may stabilize lung fibroblasts and inhabit its massive proliferation.
