糖脂平对胰岛素抵抗大鼠AMPK/PGC-1α信号通路的影响 被引量:3

摘要 目的了解中药复方糖脂平对高脂饮食诱导的胰岛素抵抗(IR)大鼠骨骼肌细胞腺苷酸活化蛋白激酶(AMPK)/过氧化物酶体增殖物激活受体γ辅助因子1α(PGC-1α)信号通路的影响。方法选择体重在180~200g的雄性SD大鼠40只,随机分为空白对照组10只和造模组30只。空白对照组全程予普通饲料喂养,造模组全程予高脂饲料喂养。8周造模成功后,将造模组再次随机分为模型组、糖脂平组、罗格列酮组,每组10只。糖脂平组予糖脂平按生药20 g/(kg·d)灌胃;罗格列酮组予罗格列酮0.8 mg/(kg·d)灌胃;模型组和空白对照组灌服等量的生理盐水,持续8周,末次给药后禁食12 h,用正常血糖高胰岛素钳夹实验评价胰岛素敏感性,检测各组大鼠体重、血糖、血胰岛素水平,并留取骨骼肌,采用Western Blot法检测AMPK-α、p-AMPK-α和PGC-1α的表达。结果中药复方糖脂平可以改善骨骼肌组织胰岛素的敏感性;降低IR大鼠体重、血糖和血胰岛素水平;糖脂平可提高IR大鼠p-AMPK-α和PGC-1α的蛋白表达。结论糖脂平具有降糖、减重、提高骨骼肌组织对胰岛素的敏感性的作用,作用机制可能与激活大鼠骨骼肌组织AMPK/PGC-1α信号通路有关。 Objective To understand the impacts of Chinese herbal compound medicine,tangzhiping on AMPK( AMP- activate protein kinase) / PGC- 1α( peroxisome proliferator activated receptor- 1α)signal pathway in the skeletal muscle of the rats of insulin resistance( IR) induced by high- fat diet. Methods Forty male SD rats,weighted from 180 g to 200 g were randomized into a blank control group( 10 rats) and a model group( 30 rats). In the blank control group,the rats were fed with common fodder. In the modeling group,the rats were red with high- fat fodder. After successful modeling in 8 weeks,the rats in the modeling group were subdivided randomly into a model group,a tangzhiping group and a rosiglitazone group,10 rats in each one. In the tangzhiping group,the lavage was done with the raw materials of tangzhiping,20 g /( kg·d).In the rosiglitazone group,the lavage was done with rosiglitazone,0. 8 mg /( kg·d). In the model group and blank control group,lavage was done with physical saline of the same dose. The lavage lasted for 8 weeks. After fasting for 12 h at the last medication,the euglycemic- hyperinsulinemic clamp technique was adopted to evaluate insulin sensitivity. The body weight,blood glucose and insulin were detected in each group. The skeletal muscle was remained. Western blot method was used to detect the expression of AMPK- α,p- AMPK-α and PGC- 1α. Results Tangzhiping,the compound herbal medicine,improved the insulin sensitivity in skeletal muscle,reduced the levels of body weight,blood glucose and insulin in IR rats and increased the proteinexpressions of p- AMPK- α and PGC- 1α. Conclusion Tangzhiping reduces the blood glucose and body weight,improves insulin sensitivity of skeletal muscle. The effects of it are possibly related to the activation on AMPK / PGC- 1α signaling pathway in the tissues of skeletal muscle.
