和厚朴酚对全脑缺血再灌注犬血浆TXB_2、6-Keto-PGF_(1α)、ET、NO的影响 被引量:11

摘要 目的观察和厚朴酚(Honokiol)对全脑缺血再灌注犬血浆B2(TXB2)、6-酮-前列腺素F1α(6-Keto-PGF1α)、内皮素(ET)、一氧化氮(NO)的影响,评价其治疗脑卒中的抗血栓作用机制。方法取杂种犬,麻醉,手术阻断左右颈总动脉、左右椎动脉,20 min后经右侧颈总动脉、椎动脉再灌注,复制犬全脑缺血再灌注损伤模型,再灌注20 min后给药。模型组静脉注射0.9%氯化钠注射液,尼莫地平组静脉注射尼莫地平15μg/(kg·h),Honokiol高、中、低剂量组分别静脉注射Honokiol40μg/kg、8μg/kg、1.5μg/kg。各组动物给药时用生理盐水将药物稀释至15 ml,10 min内静脉注射完毕。给药后2.5 h,测定血浆TXB2、6-Keto-PGF1α、ET、NO水平。结果 Honokiol高、中、低剂量组血浆中TXB2较模型组降低,其中Honokiol低剂量组与模型组比较差异有统计学意义(P<0.05),Honokiol对血浆6-Keto-PGF1α、ET、NO影响不大。结论和厚朴酚静脉注射通过降低全脑缺血再灌注犬血浆TXB2含量发挥抗血栓作用。 Objective To investigate the effect of Honokiol on TXB2、6- Keto- PGF1α、ET、NO after global cerebral ischemia- reperfusion injury in dog and to evaluate its mechanism of antithrombus in cerebral apoplexy. Methods Four vessels leading to the brain of anesthetized- dog were blocked 20 min by surgery. And then,the right internal carotid artery and right vertebral artery were released 20 min in order to copy dog global cerebral ischemia and reperfusion injury model. The model dogs were divided into five groups. Honokiol 40 μg / kg,8 μg / kg,1. 5 μg / kg was injected into intravenous of the Honokiol respectively as high-,medium- and low- dosed group. 0. 9% Na Cl and Nimodipine 15 μg / kg / h was administered respectively to model control and nimodipine group. Delivery volume was 15 ml,intravenous injection was completed within 10 min. TXB2,6- Keto- PGF1α,ET and NO in plasma was measured 2. 5 hours after administration.Results The TXB2 was reduced in each Honokiol dose- groups compared with model group. But the effects on 6- Keto- PGF1α,ET,NO was a little. Conclusion Honokiol intravenous injection can reduce TXB2 after global cerebral ischemia- reperfusion in dog.
