摘要
目的探讨天麻钩藤饮正交方不同配伍剂量对高血压大鼠的降压效应及内皮素-1(ET-1)、超氧化物歧化酶(SOD)、丙二醛(MDA)的影响。方法将50只SD大鼠随机分为10组,即对照组、模型组、正交1~8号不同剂量配方组。空白组和模型组给予等量蒸馏水灌胃,1~8号配方组分别给予不同剂量的天麻钩藤饮正交Ⅰ方。在造模基础上,观测给药前后大鼠血压变化,并测定最佳剂量配伍组大鼠给药后ET-1、SOD、MDA含量。结果正交1~8号不同剂量配方组给药后与给药前比较,收缩压均降低(P<0.01);与模型组比较,各用药组血压也均降低(P<0.05);但各用药治疗组间比较,差异无统计学意义(P>0.05)。依据降压效应确定的最佳配伍剂量组(1号配方)大鼠血清ET-1、MDA含量均降低(P<0.01)、而SOD含量升高(P<0.05),说明血压高低程度与血清ET-1、MDA含量呈正相关性、与SOD活性呈负相关性。结论天麻钩藤饮不同配伍剂量均有较好的降压疗效,而最佳配伍剂量组(天麻6 g,钩藤12 g,石决明18 g,桑寄生9 g,川牛膝9 g)可降低ET-1、MDA含量,升高SOD含量,提示其降压作用机制与抑制氧化应激反应、保护血管内皮功能有关。
Objective To explore the anti- hypertensive effect on hypertension in the rats treated with orthogonal composition of tianma gouteng yin and the impacts on ET- 1,SOD and MDA. Methods Fifty SD rats were randomized into 10 groups,named a control group,a model group,and No1. to No. 8 orthogonal composition groups. In the blank and model groups,the distilled water was used for lavage at the equal dose. In No. 1 to No. 8 orthogonal composition groups,the orthogonal I formula of tianma gouteng yin was used at different doses accordingly. On the basis of modeling,blood pressure was observed before and after medications and the levels of ET- 1,SOD and MDA were determined after medication in the optimal composition group. Results The systolic pressure was reduced in all of the orthogonal composition groups as compared with that before medication( P < 0. 01). Compared with the model group,blood pressure was reduced in every medication group( P < 0. 05). The differences were not significant statistically in comparison among the medication groups( P > 0. 05). The optimal composition group( No. 1 composition group) was determined by the anti- hypertensive effect,in which,the levels of ET- 1 and MDA in serum were all reduced( P < 0. 01) and SOD was increased( P < 0. 05). It was suggested that the severity of hypertension was positively correlated with the levels of ET- 1 and MDA and negatively with SOD activity. Conclusion Tianma gouteng yin of different doses achieves the good efficacy on hypertension. The optimal composition( rehizoma gastrodiae 6 g,uncaria 12 g,concha haliotidis 18 g,herba taxilli 9 g and radix cyathulae 9 g) reduces the levels of ET- 1 and MDA and increases SOD level. It is suggested that the anti- hypertensive effect is related to the inhibition of oxidative stress reaction and protection of vascular endothelial function.
