摘要
目的观察钩薄过敏颗粒对咳嗽变异性哮喘(CVA)大鼠肺泡灌洗液(BALF)中白细胞介素-4(IL-4)、白细胞介素-5(IL-5)、干扰素-γ(IFN-γ)水平的影响,并探讨部分作用机制。方法 60只雄性SD大鼠随机分为空白对照组,模型对照组,地塞米松组,钩薄过敏颗粒高、中、低剂量组,每组10只。除空白对照组外,其余各组利用卵蛋白(OA)和氢氧化铝共同激发致敏建立CVA大鼠模型。各组分别给予不同药物灌胃,钩薄颗粒低、中、高剂量组分别给予剂量为6.71 g/kg、13.42g/kg、28.63 g/kg的钩薄过敏颗粒,地塞米松组给予地塞米松0.5 mg/kg。空白对照组和模型对照组用蒸馏水(1 ml/100 g)。各组连续给药13 d,第14天处死动物,试剂盒检测肺泡灌洗液中IL-4、IL-5、IFN-γ的含量,HE染色观察肺组织病理学改变。结果与空白对照组比较,模型对照组BALF中IL-4、IL-5含量明显升高(P<0.05),IFN-γ含量明显降低(P<0.05);与模型对照组相比,钩薄过敏颗粒高剂量组IL-4、IL-5明显降低(P<0.05),IFN-γ含量明显升高(P<0.05)。肺组织病理观察显示,钩薄过敏颗粒可以减少CVA引起的炎细胞浸润、支气管上皮杯状细胞增多,渗出物的生成,尤其是高剂量组最为明显。结论钩薄过敏颗粒可有效治疗CVA大鼠,其机制可能通过调节IL-4、IL-5、IFN-γ的含量并减轻炎性细胞浸润有关。
Objective To observe impacts of goubo guomin granules on IL- 4,IL- 5 and IFN- γin bronchoalveolar lavage fluid( BALF) in the rats of cough variant asthma( CVA) and explore the partial mechanism. Methods Sixty male SD rats were randomized into a blank control group,a model control group,a dexamethasone( DXM) group,the high- dose,middle- dose and low- dose groups of goubo guomin granules,10 rats in each one. Except the blank control group,OA and AL( OH) 3 were used to induce CVA rat models. Lavage was done with different drugs. The doses were 6. 71 g/kg,13. 42 g/kg and 28. 63 g/kg for goubo guomin granules in the high- dose,middle- dose and low- dose groups separately. DXM,0. 5mg/kg was used in the DXM group. The distilled water of the same dose was used in the blank control group and the model control group. The medication was given continuously for 13 days in each group. On the 14 th day,the rats were sacrificed. The kit was used to determine the levels of IL- 4,IL- 5 and IFN- γ in BALF. HE-staining method was used to observe the pathological changes of lung tissue. Results Compared with the blank control group,the levels of IL- 4 and IL- 5 were increased apparently in BALF of the model control group( P < 0. 05) and the level of IFN- γ was reduced apparently( P < 0. 05). Compared with the model control group,in the high- dose group,the levels of IL- 4 and IL- 5 were reduced apparently in the model control group( P < 0. 05) and the level of IFN- γ was increased apparently( P < 0. 05). The pathological observation showed that goubo guomin granules reduced inflammatory cell infiltration induced by CVA,increased bronchial epithelial goblet cell and induced exudates,which is the most apparent in the high- dose group. Conclusion Goubo guomin granules treat effectively CVA in the rats possibly through the regulation of IL- 4,IL-5 and IFN- γ and the alleviation of inflammatory cell infiltration.
