龙葵正丁醇提取物抗人结直肠癌SW480细胞增殖及其作用机制研究

Study of Solanum nigrum n-butanol extract on the proliferation of human colorectal cancer SW480 and its mechanism

目的探讨龙葵正丁醇提取物对人结直肠癌SW480增殖抑制作用及其作用机制。方法采用MTT法测定细胞增殖抑制率,流式细胞仪分析细胞周期的变化,比色法检测细胞内Caspase-3活性改变。结果龙葵正丁醇提取物对人结直肠癌SW480细胞有明显的生长抑制作用,并呈现剂量依赖性。经龙葵正丁醇提取物作用后,SW480细胞周期显著改变,出现G2/M期阻滞,Caspase-3蛋白表达升高。结论龙葵正丁醇提取物能抑制人结直肠癌SW480增殖。上调Caspase-3蛋白的表达,改变细胞周期可能是其机制之一。

Objective To investigate the inhibitory effect of Solanum nigrum n-butanol extract on the proliferation of human colorectal cancer SW480 and its mechanism.Methods MTT assay was used to determine the inhibition rate of cell proliferation.Flow cytometry was used to analyze the changes of cell cycle.the activity of Caspase-3 was detected by colorimetry.Results The n-butanol extract of Solanum nigrum could inhibit the growth of human colorectal cancer SW480 cells in a dose-dependent manner.After treated with Solanum nigrum n-butanol extract,the cell cycle of SW480 was significantly changed,G2/M phase arrest was observed,and the expression of Caspase-3 protein was increased.Conclusion The butanol extract of Solanum nigrum can inhibit the proliferation of human colorectal cancer SW480 and up-regulation of Caspase-3 protein expression.Alteration of cell cycle may be one of its mechanisms.
