摘要
优化了枯草芽孢杆菌KD-N2利用茶籽粕等为底物液体发酵生产蛋白酶的发酵条件。单因素实验结果表明,在初始p H 8.0、种龄30h、转速230r/min、接种量12%(v/v)、培养温度28℃、装液量30m L时,发酵液中蛋白酶活性较高。正交试验结果表明,接种量5%(v/v),装液量30m L,初始p H值7.2时为最佳条件,在30℃、30h种龄、200r/min培养18h后蛋白酶活性达到104.2U。文章同时对发酵过程可溶性蛋白质含量和发酵液p H值变化进行了分析,为提高发酵产酶总量,发酵条件还需进一步进行优化。
Fermentation conditions for proteinase production on tea seed as the substrate by Bacillus subtilis KD-N2 were optimized. Single factor design experiments results showed that initial p H value of 8.0, seed age of 30 h, rotation speed of 230r/min, inoculum of 12%, culture temperature of 28℃, 30 h, culture volume of 30 m L in 250 m L flasks. Orthogonal experiments showed that the optimal conditions were of initial p H value 7.2, inoculum 5%(v/v),culture volume 30 m L, after cultured at 30℃, 200r/min with 30 h seed age for 18 h, the proteinase activity reached 104.2U. The soluble protein contents as well as culture p H value were also analyzed in this study, culture conditions should be further optimized for proteinase activity improvement..
