摘要 对棘孢小单孢菌产庆大霉素B发酵培养基进行优化,应用Design Expert 7.0软件设计Plackett-Burman筛选实验,筛选出硝酸钾、黄豆饼粉及氯化钴三个显著影响因素,通过正交实验对其进一步优化。确定最优的发酵培养基配方为:玉米淀粉6%,葡萄糖0.5%,玉米蛋白粉2.25%,黄豆饼粉3.5%,蛋白胨0.3%,鱼粉0.1%,硝酸钾0.02%,硫酸铵0.075%,碳酸钙0.6%,氯化钴0.001%,庆大霉素B效价可达800U/m L,比原配方提高20%。 The media for gentamicin B production excreted by Micromonospora echinospora was optimized by Plackett-Burman and orthogonal experiment. Firstly, Plackett-Burman experiment was carried out and the result was analysed by software design expert 7.0, through which three components potassium nitrate, cornstarch and cobalt chloride were confirmed as significant factors. Further, the three components were optimized using orthogonal experiment and the results showed that the optimal culture media was as follows:cornstarch 6%, glucose 0.5%, corn gluten meal 2.25%, soybean meal 3.5%, peptone 0.3%, fish meal 0.1%, potassium nitrate 0.02%, ammonium sulfate 0.075%, calcium carbonate 0.6%, cobalt chloride 0.001%. The yield of gentamicin B was increased about 20% compared with control media and the potency can reach 800U/m L.
