摘要
线粒体ATP依赖的LON蛋白水解酶(又称LONP1),是由核基因LONP1编码的线粒体的重要蛋白之一,在调节线粒体代谢活动、维持线粒体稳态和修复线粒体DNA中具有重要作用。近年来,有关LONP1参与的生理功能以及人类疾病(包括癌症、神经退行性疾病、心脏病和中风)的研究逐渐增加,研究者已经明确,许多线粒体疾病的发生发展机制涉及不同程度的LONP1功能障碍。本文对LONP1的正常功能、在恶性肿瘤细胞中的表达情况、作用机制及其对多种肿瘤发生发展的影响进行综述,旨在为临床恶性肿瘤生物学诊治提供新的靶标。
Mitochondrial ATP-dependent LON proteolytic enzyme(also known as LONP1)is one of the most important proteins encoded by the nuclear gene LONP1.It plays a vital role in regulating mitochondrial metabolic activity,maintaining mitochondrial homeostasis and repairing mtDNA.In recent years,researches on the physiological functions involved in LONP1 and human diseases(including cancer,neurodegenerative diseases,heart disease and stroke)have gradually increased,and it has been clarified that many mitochondrial diseases are involved with LONP1 dysfunction.This review summarizes the normal function of LONP1,its expression in malignant tumors,its mechanism and effects on the development of tumors,aiming to provide a new target for the clinical diagnosis and treatment of malignant tumors.
