气相色谱法多重检验海豹油中二十碳五烯酸、二十二碳五烯酸和二十二碳六烯酸方法研究 被引量:4

摘要 目的建立多重检验海豹油中二十碳五烯酸(eicosapentaenoic acid,EPA)、二十二碳五烯酸(docosapentenoic acid,DPA)和二十二碳六烯酸(docosahexaenoic acid,DHA)的气相色谱法。方法用氢氧化钾甲醇溶液将样品中EPA、DPA和DHA甲酯化,气相色谱程序升温同时分析3个ω-3多不饱和脂肪酸,分别用外标法和内标法进行定量。结果 3个ω-3多不饱和脂肪酸的外标法定量线性范围为50~5000μg/m L,相关系数均】0.999,精密度相对标准偏差均【10%,回收率均】90%;内标法定量的精密度相对标准偏差均【10%,回收率均】85%;外标法和内标法对3个ω-3多不饱和脂肪酸的结果比较,CV%均小于10%。结论该方法操作简单快捷,适用于进口海豹油中EPA、DPA和DHA的含量简便和准确的测定。 Objective To establish a method for the determination of eicosapentaenoic acid (EPA), docosapentenoic acid (DPA), and docosahexaenoic acid (DHA) in seal oil by gas chromatography. Methods The EPA, DPA, DHA in the samples were methylated with KOH-CH3OH solution. Three kinds of ω-3 polyunsaturated fatty acids were analyzed simultaneously by using temperature rising program of gas chromatograph, and quantified by the external standard method and internal standard method. Results The linear range of the external standard of 3 kinds of ω-3 polyunsaturated fatty acids were 50~5000 μg/mL, the correlation coefficients were all over 0.999, the relative standard deviations (RSDs) of precision were less than 10%, and the recovery rates were all over 90%. RSDs of precision of internal standard method were lower than 10% and the recovery rates were all over 85%. The CV% of external standard method and internal standard method for the 3 kinds of ω-3 polyunsaturated fatty acids were less than 10%. Conclusion This method is simple and rapid, and can be used to determine the contents of DHA, DPA and EPA in imported seal oil easily and accurately.
