摘要
目的建立一种反相高效液相色谱柱前在线衍生法测定保健品中氨基葡萄糖硫酸钾盐的含量。方法利用邻苯二甲醛衍生试剂与氨基葡萄糖硫酸钾盐中的氨基进行柱前在线衍生化反应,采用C18色谱柱分离,以0.68%醋酸缓冲液:甲醇(V:V=82:18)为流动相,流速1.0 m L/min,检测波长340 nm。结果 D-盐酸氨基葡萄糖浓度在0.052~1.045 mg/m L之间呈现良好的线性,平均回收率为97.5%,RSD%=1.2(n=9)。结论该方法推广性高、简便、准确,可用于制剂及保健品中氨基葡萄糖硫酸钾盐的含量测定。
Objective To establish a method for the determination of N-Sulfo-glucosamine potassium salt by reversed-phase high performance liquid chromatography (RP-HPLC) with online pre-column derivation in health food. Methods Online pre-column derivation were reacted by using phthalic dicarboxaldehyde (OPA) derived reagents and amino of N-Sulfo-glucosamine potassium salt. The products were separated by C18 column with 0.68%acetic acid buffer:methanol (V:V=82:18) as mobile phase, the flow rate of 1.0 mL/min, and detection wavelength of 340 nm. Results D-glucosamine hydrochloride concentration had a good linearity in the range of 0.052~1.045 mg/mL. The average recovery rate was 97.5%, RSD%=1.2 (n=9). Conclusion The method is high, simple and accurate, and can be used to determine the content of N-Sulfo-glucosamine potassium salt in pharmaceutics and health food.
