Determination of inositol in functional drink by high performance liquid chromatography-tandem mass spectrometry

摘要 目的建立液相色谱-串联质谱法测定功能饮料中肌醇的含量。方法功能饮料振荡混匀后取样0.2mL,用50%的乙腈水稀释500倍,过0.22μm的微孔滤膜后直接进样,经BEH Amide色谱柱(2.1mm×100 mm, 1.7μm)洗脱,流动相为乙腈-水,柱温为30℃,流速为0.3 mL/min,用串联四极杆质谱采用多反应监测模式进行测定,外标曲线法定量。结果肌醇在0.05~2.0μg/mL范围内线性关系良好,线性相关系数r为0.9999,检出限和定量限分别为0.65 mg/L和2.15 mg/L;加标浓度为0.025、0.125和0.25 mg/mL的平均回收率分别为103.5%、100.0%和101.8%,批内精密度分别为3.3%、1.1%和1.5%,批间精密度分别为3.3%、2.0%和1.9%。结论该方法前处理简单、快速、准确、灵敏,可以满足实际检测分析的需要。 Objective To establish a method for the determination of inositol in functional drink by high performance liquid chromatography-tandem mass spectrometry. Methods After shaking and mixing, 0.2 mL functional drink was diluted 500 times with 50% acetonitrile water, and directly injected after passing through 0.22 μm microporous membrane filter. Inositol was separated by BEH Amide column(2.1 mm×100 mm, 1.7 μm) with acetonitrile-water as the mobile phase at 30 ℃, and the flow rate was 0.3 mL/min. The sample was determined by tandem quadrupole mass spectrometry using multi-reaction monitoring mode, and quantified by external standard method. Results The inositol had a good linear relationship in the concentration range of 0.05-2.0 μg/mL with correlation coefficients of 0.9999. The limit of detection and limit of quantitative were 0.65 mg/L and 2.15 mg/L, respectively. The average recovery of inositol with the spiked concentration of 0.025, 0.125 and 0.25 mg/mL were 103.5%, 100.0% and 101.8%, the intra-batch precision were 3.3%, 1.1% and 1.5%, and the inter-batch precision were 3.3%、2.0%和1.9%, respectively. Conclusion This method is simple, accurate and sensitive, which can meet the requirements of practical analysis.
