原花青素含量测定方法考察及应用 被引量:11

摘要 以慕萨莱思酒为原料,对3种原花青素测定方法 (香草醛-盐酸法、香草醛-硫酸法、正丁醇-盐酸法)进行方法学考察,筛选出葡萄酒原花青素测定方法,并对7种葡萄酒原花青素含量进行比较。对3种方法进行精密度试验、方法回收率试验和稳定性试验。选用优化方法对7种葡萄酒原花青素进行含量测定。结果表明, 3种方法精密度分别为6.23%, 9.87%和19.12%,加标回收率分别为15.23%, 48.44%和69.77%,稳定性从高到低依次为:盐酸-香草醛法、硫酸-香草醛法、盐酸-正丁醇法,其中盐酸-香草醛法有较高精密度、稳定性,而回收率严重偏低,原因有待考察。用该方法测定7种葡萄酒原花青素含量并加以比较,长城干红葡萄酒、张裕红红葡萄酒、威龙红葡萄酒、慕萨莱思中原花青素含量较高,分别为1 100.80, 996.5, 344.3和330.8μg/mL。 With moussa rice wine as raw material,the three methods of determination of procyanidins(vanillin-hydrochloric acid method,vanillin-sulfuric acid method,and n-butyl alcohol and hydrochloric acid method)were investigated,and the wine procyanidins determination method was selected,to compare seven kinds of wine procyanidins content.The three methods of precision,the recovery and stability were tested and the optimal method was selected to determine the procyanidins content of7 kinds of wines.Results show that the precisions of three methods are 6.23%,9.87%and 19.12%,respectively;The standard addition recovery rates are 15.23%,48.44%and 69.77%,respectively;The hydrochloric acid-vanillin method had higher precision and stability,but seriously low recovery rate,with no founding reason.The procyanidins content of 7 kinds of wines was determined and compared,and the Great Wall brand dry red wine,Zhang Yugong red wine,the veyron red wine and moussa had comparatively higher anthocyanin content,which were 1 100.80,996.5,344.3 and 330.8μg/mL,respectively.
