使用高效液相色谱,建立一种定量测定干生姜中姜黄素类化合物含量的方法,并对五种不同地区的干生姜和三种姜黄素产品进行含量分析。色谱条件:流动相为V (乙腈)︰V (水)=50︰50 (用磷酸调节pH 3),流速0.8 mL/min,柱温35℃,采用紫外检测器(UV)检测,检测波长425 nm。方法学考察表明,三种姜黄素在0.5~100μg/m L范围内线性关系良好,相关系数为0.999 7,最低检出限0.01μg/mL,在10~100 mg/kg加标浓度范围内平均回收率分别为双去甲氧基姜黄素100.59%、去甲氧基姜黄素100.95%、姜黄素96.86%。五个地区(广东、广西、四川、云南和安徽)干生姜中姜黄素总含量分别为3.03, 1.42, 0.90, 2.25和6.23 mg/kg。对三个市销产品进行检测,结果表明检测方法稳定、可靠,可应用于检测实际样品。
A method for the quantitative determination of curcumin in dried ginger was established by high performance liquid chromatography(RP-HPLC).Contents of dried ginger in five different regions and three kind of curcumin products were analyzed.Chromatographic conditions were that the mobile phase was V(acetonitrile)︰V(water)=50︰50(pH 3,adjusted with phosphoric acid),the flow rate was 0.8 mL/min,the column temperature was 35℃,and the UV lamp wavelength was425 nm.Methodological examination shows that the three samples of curcumin in the range of 0.5 to 100μg/mL were good,the correlation coefficient was 0.999 7,and the minimum detection limit was 0.01μg/mL.Within the spiked concentration range of 10 to 100 mg/kg,the average recoveries are 100.59%of double demethoxy curcumin,100.95%of demethoxy-curcumin,and 96.86%of curcumin.The total contents of curcumin in dried ginger in five areas(Guangdong,Guangxi,Sichuan,Yunnan and Anhui)were 3.03,1.42,0.90,2.25 and 6.23 mg/kg,respectively.The three marketed products were tested,and the detection method was stable and reliable.It could be used to detect actual samples.
