摘要
建立桂花中6种单糖的含量测定方法。采用PMP柱前衍生法和高效液相色谱法(HPLC)分离检测6种单糖含量的方法。色谱条件:色谱柱为Eurosp Her 100-5 C_(18) (200 mm×4.6 mm, 5μm),流动相为乙腈-磷酸缓冲液(18∶82),柱温40℃,流速1.0 mL/min,检测波长250 nm,进样量20μL。所建立的PMP衍生化-HPLC法可准确地测定桂花中甘露糖(Man)、葡萄糖(Glu)、鼠李糖(Rha)、木糖(Xyl)、果糖(Fru)、半乳糖(Gal)这6种单糖。结果表明:HPLC柱前衍生法测定单糖含量简便、快捷,并且测定结果相差较小,灵敏度高,测定结果准确,重现性好。
Research established the determination methods of six monosaccharides for Osmanthus fragrans.Method for the separation and detection of six monosaccharides is using PMP precolumn derivatization and high performance liquid chromatography(HPLC).Chromatographic conditions are that the column is Eurosp Her 100-5 C18(200 mm×4.6 mm,5μm).The mobile phase is acetonitrile-phosphate buffer(18∶82),column temperature is 40℃,flow rate is 1.0 mL/min,detection wavelength is 250 nm,and injection volume is 20μL.The established PMP derivatization-HPLC method can accurately determine the six sugars of mannose,glucose,rhamnose,xylose,fructose and galactose in sweet-scented Osmanthus fritillaria.The results showed that HPLC precolumn derivatization method was simple and rapid in determination of monosaccharide content,and the data showed little difference,high sensitivity,accurate measurement results and good reproducibility.
