HPLC Determination of Cyanidin-3-O-Glucoside Content in Different Solvent Extraction from Blueberry

摘要 建立以高效液相色谱法测定3种不同溶剂提取蓝莓花青素中矢车菊素-3-O-葡萄糖苷的含量。采用Agilent ZORBAX SB-C18色谱柱(150 mm×4.60 mm, 5μm);流动相, A相为乙腈, B相为1.5%甲酸。流速1.0 mL/min,柱温30℃,检测波长520 nm,进样量10μL。结果表明,矢车菊素-3-O-葡萄糖苷在9.756~48.78μg/mL范围内(R2=0.999 1)线性关系良好,最低检出限为0.06 mg/kg,精密度n为6组, RSD=1.36%。3种不同溶剂平均回收率分别为101.67%, RSD=3.97%;96.55%, RSD=1.05%;98.28%, RSD=2.58%。不同溶剂提取蓝莓花青素中矢车菊素-3-O-葡萄糖苷含量不同。该方法简便、快速、重复性好,可用于蓝莓花青素的质量控制。 To establish a HPLC method for determining the contents of cyanidin-3-O-glucoside(C3 G)content in blueberry,HPLC was carried out on an Agilent ZORBAX SB-C18(150 mm×4.60 mm,5μm),with the mobile phase of A(acetonitrile)-B(1.5%formic acid),the flow rate of 1.0 mL/min,the column temperature at 30℃,the detection wavelength of 520 nm,and the sample size of 10μL.The mobile phase was gradient elution.The C3 G was in good linearity within the range of 9.756-48.78μg/m L(R2=0.999 1).The method of MDL was 0.06 mg/kg.The RSD of the precision of the method was 1.36%with N=6;The average recoveries of three solvents were 101.67%,RSD=3.97%;96.55%,RSD=1.05%;98.28%,RSD=2.58%.Different solvent extraction of C3 G content in blueberry anthocyanins was different.The method was simple,fast and reproducible,and could be applied for quality control of blueberry.
