摘要
采用原子吸收光谱法测定由糯米、粳米、家蚕蛹浸提液培养出来的冬虫夏草中的铁、锌的含量。采用干法灰化样品, 650℃灰化。用硝酸溶解灰分。依据测定的吸光度与浓度建立工作曲线,计算出样品中铁、锌的含量。加标中浓度采用低、中、高3档,分别是0.10, 0.50和1.00 mg/kg。测定结果表明,糯米、粳米、家蚕蛹浸提液培养出的冬虫夏草中的铁含量分别为26.9, 49.08和35.16 mg/kg;糯米、粳米、家蚕蛹浸提液培养出的冬虫夏草中的锌的含量为13.28, 22.47和18.90 mg/kg。6份样品3组平行样的相对标准偏差为0.025%~0.059%,最低检出限为0.2 mg/kg,加标回收率为94.27%~98.77%。该方法检测度高、线性范围好、灵敏度高,对检测生物样品有一定的成效,适用于检测大量的生物样品。
The content of iron and zinc in Cordyceps sinensis cultured from glutinous rice,japonica rice and silkworm chrysalis was determined by atomic absorption spectrometry.Using dry ashing sample,650℃ashing,the ash was dissolved in nitric acid.Based on the determination of absorbance and concentration to establish a working curve,the content of iron and zinc of the sample was calculated.The standard concentration was 0.10,0.50 and 1.00 mg/kg.The results were as follows:the iron contents of Cordyceps sinensis cultured from the extracts of glutinous rice,japonica rice and domestic silkworm chrysalis were 26.98,49.08 and 35.16 mg/kg,respectively.The contents of zinc in Cordyceps sinensis cultured from the extracts of glutinous rice,japonica rice and silkworm chrysalis were 13.28,22.47 and 18.90 mg/kg,respectively.The relative standard deviation of three groups of six samples was 0.025%-0.059%;The minimum detection limit was 0.2 mg/kg,and the standard recovery was 94.27%-98.77%.The method had high detection degree,good linear range and high sensitivity,and it was effective in detecting biological samples.
