摘要
建立液相色谱法测定辣木叶壳寡糖袋泡茶中壳寡糖的含量。用水提取,6.0 mol/L的盐酸溶液水解辣木叶壳寡糖袋泡茶中的壳寡糖,使其水解为D-盐酸氨基葡萄糖,利用液相色谱仪作为检测手段,紫外检测器进行定性定量分析。本文通过对方法的系统适用性、专属性、精密度、回收率、线性范围及耐用性方面的考察来建立辣木叶壳寡糖袋泡茶中壳寡糖的含量测定的方法,重复性实验的相对标准偏差为0.15%,回收率为94.52%~98.52%,在进样量0.091 3~0.913μg内呈线性,r=0.998 68,符合要求。
Determination method was developed for the content of chitosan oligosaccharide of tea in Moringa leaves chitosan tea by high performance liquid chromatography(HPLC). The samples were extracted by water, chitosan oligosaccharide was hydrolyzed to D-glucosamine hydrochloride by 0.6 mol/L hydrochloric acid, high performance liquid chromatography(HPLC) was the method of determination, UV as the detector for qualitative analysis. This thesis investigated the method from system suitability, specificity, precision, recovery percent, linear range and durability. Relative standard deviation of the repetitive experiments was 0.15%, the recovery percent was 94.52%~ 98.52%, the determination method provided a good linearity from 0.0913 μg to 0.913 μg,r=0.998 68. The result conformance to requirements.
