蜂胶中Artepillin C的高效液相色谱法测定

Determination of Artepillin C in Propolis by High Performance Liquid Chromatography

建立了利用高效液相色谱法测定蜂胶中Artepillin C的方法,优化了萃取剂对萃取效率的影响.实验测试表明,80%甲醇提取效果最佳,采用Venusil MP C18柱(4.6 mm×250 mm,5μm),以甲醇和0.4%磷酸(V/V)水溶液作为流动相进行梯度淋洗,检测波长312nm,流速1.0mL/min,柱温30℃的色谱条件,可实现Artepillin C的分离检测.该方法样品前处理简单快捷,操作方便,检出拥有良好的线性关系,线性方程为y=18 944 500 x-2 385(R2=1.000 0),检出限(0.20 mg/L),回收率(95.0%~99.4%)和重现性(RSD≤1.01%,n=6).

A method for determining Artepillin C is developed using high performance liquid chromatography.In this study,extraction agents are optimized and the results show that 80% methanol can achieved the best extraction rate.The chromatographic separation is performed on a Venusil MP C18column(4.6mm×250mm,diameter 5μm).Mobile phase is the mixture of methanol and 0.4 % orthophosphoric acid(V/V)for gradient elution and the flow rate is 1.0mL/min.The operating temperature of the column is set at 30 ℃ and the detection wavelength(UV)is 312 nm.Further experiments have showed that the method has good linear relationship(R2=1.000 0),low detection limit(0.20mg/L),high recovery(95.0%~99.4%)and good precision(RSD≤1.01%,n=6).This method is suitable for the determination of Artepillin C in propolis sample.
