摘要
文章建立阴离子交换色谱—积分脉冲安培法检测5种低聚壳寡糖(壳二糖、壳三糖、壳四糖、壳五糖、壳六糖)的方法.采用色谱柱为CarboPac PA10分析柱(4.0×250mm),柱温30℃;淋洗液为15mmol/L NaOH溶液,等度淋洗,流速0.4mL/min;进样量20μL.结果表明:5种低聚壳寡糖的质量浓度0.05~2.00μg/mL范围内与色谱峰面积呈良好线性关系(r2=0.995 9~0.999 8).平均回收率(n=3)在90.85%~114.21%,RSD<3.0%.方法可快速简便地测定市售壳寡糖产品中5种低聚壳寡糖的含量.
An anion exchange chromatography with integrated pulsed amperometric detection(AEC-IPAD)method was developed for the simultaneous analysis of five small chitooligosaccharides(Chitobiose,Chitobiose,Chitotetraose,Chitopentaose,Chitohexaose).Separation was performed on CarboPac PA10 ion exchange column(Dinex,4.0×250mm)and column temperature was 30°C.The mobile phase was 15mmol/L aqueous sodium hydroxide with isocratic elution detection at a flow rate of 0.4mL/min and injection volume of 20μL.Results showed that the method exhibited a good linearity(r2=0.995 9~0.999 8)in the ranges of 0.05~2.00μg/mL.The average recoveries(n=3)of the five chitooligosaccharides were from 90.85% to 114.21% and RSDs of recoveries were less than 3.0%.The method can be applied as a rapid and convenient evaluation method for the quality of chitooligosaccharide products that is commercially available.
