刺玫果提取物抗衰老作用的研究 被引量:11

Anti-aging effect of Rosa davurica Pall extract on Drosophila melanogaster

摘要 将野生刺玫果用80%乙醇回流提取,冻干成粉,测定总黄酮含量。在培养基中添加低(0.115%)、中(0.461%)、高(1.845%)不同浓度的刺玫果冻干粉喂养果蝇,对果蝇寿命及寿命相关行为学指标进行考察。研究发现刺玫果提取物的总黄酮含量为220.6 mg/g。在果蝇寿命实验中,当给药剂量为中(0.461%)剂量时,雌雄果蝇的平均寿命达到最高。高(1.845%)、低(0.115%)剂量也对果蝇寿命有一定的延长作用,雌雄果蝇寿命之间存在一定的差异。在抗亚急性氧化损伤实验中,与空白组相比雌果蝇在中剂量组(0.461%)存活时间最长,延长27.40%,而雄果蝇在高剂量组(1.845%)存活时间最长,延长28.65%。此结果同样体现在30日龄果蝇的攀爬实验和体内水分含量测定实验中。在果蝇繁殖实验显示,低剂量组(0.115%)7、15日龄的果蝇在7 d内出生的子代数量最多,中剂量组(0.461%)30 d的果蝇在同期出生的果蝇子代数量最多,而高剂量组(1.845%)呈现抑制作用。结果表明,刺玫果提取物具有抗衰老作用,可延缓果蝇寿命、改善果蝇寿命相关的行为学指标。 To study the anti-aging effect of Rosa davurica Pall extract on Drosophila melanogaster. 80%ethanol reflux extraction was used on wild Rosa davurica Pall and freeze-dried it into powder to determine total flavonoids. Drosophila melanogaster were fed with the freeze-dried powder of Rosa davurica Pall with low(0.115%), medium(0.461%) and high(1.845%) concentrations in culture medium. The life span and the related behavior indexes of life span of Drosophila melanogaster were investigated. The total flavonoid content of Rosa davurica Pall extract is 220.6 mg/g. In the life span test of Drosophila melanogaster, the average life span of female and male Drosophila melanogaster could reach the highest when the dose was medium(0.461%). The life span of Drosophila melanogaster was also prolonged with the high dose of 1.845% and the low dose of 0.115%, and there were some differences between male and female Drosophila melanogaster. In the anti-subacute oxidation injury experiment, female Drosophila melanogaster had the longest survival time in the medium dose group(0.461%) compared with the blank group, which prolonged 27.40%, while the male Drosophila melanogaster had the longest survival time in the high dose group(1.845%), which prolonged 28.65%. The results were also reflected in the climbing experiment and the determination of water content in the body of the 30-day-old Drosophila melanogaster.In the reproduction experiment of drosophila melanogaster, Drosophila melanogaster aged 7 days to15 days in the low dose group(0.115%) had the highest number of offspring born within 7 days, and the30-day-old Drosophila melanogaster in the medium dose group(0.461%) had the largest number of offspring born at the same time, while the high dose group(1.845%) presented the inhibitory effect. The Rosa davurica Pall extract has anti-aging effect, which can delay the life span of Drosophila melanogaster and improve the behavior indexes related to the life span of Drosophila melanogaster.
