美洲合欢花中花色苷的提取工艺初探及其抗氧化活性研究 被引量:4

摘要 采用乙醇回流法提取美洲合欢花中的花色苷,以花色苷提取量为考察指标,考察了料液比、提取温度、pH、提取时间和乙醇浓度对花色苷提取效率的影响。在单因素试验基础上,通过正交试验优化美洲合欢花花色苷的提取工艺。结果表明,在提取时间为60 min、提取溶剂pH值为2.0的条件下,花色苷最佳提取条件为:料液比1:70(g/mL)、提取温度60℃、乙醇浓度60%,提取量为7.29 mg/g。通过测定最佳工艺下美洲合欢花花色苷提取物对DPPH自由基、羟自由基和ABTS^+·自由基清除能力,考察美洲合欢花花色苷抗氧化活性。结果表明,美洲合欢花花色苷对DPPH自由基和羟自由基清除能力优于Vc,而对ABTS+·自由基稍弱于Vc。 The anthocyanins extracted from Calliandra haematocephala were extracted by ethanol refluxing method. The anthocyanin extraction amount was used as an index to examine the effect of the ratio of material to liquid, extraction temperature, pH, extraction time, and ethanol concentration on the extraction efficiency of anthocyanins. Above, the extraction process of anthocyanin from Calliandra haematocephala was optimized by orthogonal test. The results showed that under the conditions of 60 min extraction time and pH 2.0 of the extraction solvent, the optimum extraction conditions of anthocyanin from Calliandra haematocephala were: the ratio of solid to liquid 1:70(g/mL), extraction temperature 60 ℃, and ethanol volume fraction 60%, the extraction amount was 7.29 mg/g. The antioxidant activity of anthocyanins from Calliandra haematocephala was investigated by determining DPPH free radical scavenging rate, ABTS^+· free radical scavenging ability and hydroxyl radical scavenging under optimal conditions. The results showed that the scavenging ability of anthocyanin on DPPH radical and hydroxyl radical was better than that on Vc, but slightly weaker on ABTS+· free radical.
