九头狮子草提取物在模型体系中对丙烯酰胺的抑制作用研究

摘要 目的:研究九头狮子草各提取物对丙烯酰胺的抑制规律和量效关系,筛选出对丙烯酰胺的最佳抑制部位。方法:利用低湿焙烤模式下葡萄糖/天冬酰胺模型体系研究九头狮子草各提取物对丙烯酰胺的抑制作用,根据活性追踪法筛选最佳部位。结果:各提取物浓度与丙烯酰胺抑制率呈非线性关系,丙烯酰胺抑制率最高的正丁醇萃取物在添加量为3 mg/mL时抑制率为56.78%;正丁醇萃取物经大孔树脂分离所得的乙醇洗脱物浓度与丙烯酰胺抑制率亦呈非线性关系,95%、60%乙醇洗脱物对丙烯酰胺均有更好的抑制效果,最佳抑制率为95%乙醇洗脱物,添加量为5 mg/mL时抑制率为64.20%,30%乙醇洗脱物抑制效果下降。结论:九头狮子草各提取物均有丙烯酰胺抑制效果,且大多数具有较好的效果,值得作为植物功能色素开发利用。 Objective:To study the inhibition regular pattern and dose-effect relationship of extracts of Herba Peristrophe japonica on acrylamide,and to select the best inhibitory extract fraction to acrylamide.Methods:The inhibitory effect of extracts of Herba Peristrophe japonica on acrylamide was studied through the glucose/asparagine model system under the Low-wet roasting mode.The optimum extract fraction was screened by activity tracking method.Results:There was a non-linear relationship between the concentration of each extract and the inhibition rate of acrylamide.The inhibition rate of n-butanol extract with the highest inhibition rate of acrylamide was 56.78%at the dosage of 3 mg/mL.And there were also non-linear with the different concentration ethanol eluents and the inhibition rate of acrylamide,which had been separated from n-butanol extract by macroporous resin.The 95%and 60%ethanol eluents had better inhibition effect on acrylamide but the inhibition effect of 30%ethanol eluent decreased.The optimum inhibition rate was 64.20%of 95%ethanol eluent under the dosage was 5 mg/mL.Conclusion:Each extract of Herba Peristrophe japonica had inhibitory effect on acrylamide,and most of the extracts had good inhibitory effect on it,which was worth exploiting and utilizing as a plant functional pigment.
