高效液相色谱法和量子点免疫荧光层析法测定小麦及其制品呕吐毒素含量的比较 被引量:2

Determination of deoxynivalenol content in wheat and its products by HPLC and quantum dot immuneofluorescence chromatography

摘要 采用高效液相色谱法和量子点免疫荧光层析法,测定小麦及其制品中呕吐毒素(DON)含量。2种检测方法加标回收率分别为90.75%~95.85%及82.55%~98.74%,符合GB/T 27404—2008附录F中对回收率的要求;2种方法RSD分别为0.96%和3.3%,均小于5%,精密度符合定量分析要求;标准质控样的检测结果均在质控样有效值范围内,检测结果合格。对2种方法检测样品中呕吐毒素含量的结果进行T检验,结果表明检测结果无显著性差异,确定了量子点免疫荧光层析法对呕吐毒素快速定量测定的可行性及可靠性。结合2种方法的优缺点,将其同时应用到小麦等粮食中呕吐毒素的检测,将有效提高检测的效率和安全监控水平。 The contents of deoxynivalenol(DON)in wheat and its products were determined by high performance liquid chromatography and quantum dot immunofluorescence chromatography.The tagged recovery rates obtained by the two methods were 90.75%~95.85%and 82.55%~98.74%,respectively,and both of them met the recovery requirements in appendix F of GB/T 27404—2008.RSD values from the two methods were 0.96%and 3.3%,respectively.Both of them were less than 5%,and the precision met the requirements of quantitative analysis.The results from standard quality control samples were within the effective range,and the test results were qualified.T test was carried out on the results of the two methods for the determination of deoxynivalenol content in samples.The results showed that there was no significant difference in the detection results,and the feasibility and reliability of the rapid quantitative determination of deoxynivalenol by quantum dot immunofluorescence chromatography were confirmed.Therefore,the detection efficiency and safety monitoring level will be effectively improved by combining the advantages and disadvantages of these two methods,and applying them into the detection of deoxynivalenol in wheat and other grains.
