摘要 以卡拉胶为诱导物使解淀粉枯草芽孢杆菌进行固体发酵,产生卡拉胶降解酶。通过降解产物粘度的变化来确定适宜的发酵条件及降解条件。结果表明发酵物中的液体种子接种量为2%,卡拉胶的终浓度为0.1%,降解温度为50℃,pH=5,加酶量为0.5:1(w/w,酶:底物)时,此酶的降解效果最好。卡拉胶分子量由最初的1296kDa降至1660Da。 Carrageenase were produced by Bacillus amyloliquefaciens,which fermented with carrageen as inductor.The optimal condition of zymosis and degradation determined by viscosity change was achieved:liquid seed inoculate 2%,carrageen terminal concentration was 0.1%in the solid leavening,and degrading temperature of 50℃,pH 5,the enzyme quantity was 0.5:1(w/w).Its molecular weight came down from1296kDa to 1660Da.
