胰酪胨大豆琼脂培养基平皿暴露时长研究

摘要 对制药企业测定洁净区环境中沉降菌采用的胰酪胨大豆琼脂培养基平皿(简称TSA平皿)暴露后微生物生长情况进行评价,使用2种规格、3个厂家、2个时间段的方法进行对比试验。结果表明:直径为90mm,装量为30m L的TSA平皿在暴露2.1h和4.1h后,接种于表面的金黄色葡萄球菌、铜绿假单胞菌、枯草芽孢杆菌、白色念珠菌和黑曲霉的生长均符合要求;直径为90mm、装量为25m L的TSA平皿在暴露2.1h后微生物的生长符合要求,但暴露4.1h后,接种的黑曲霉生长情况不好。由此表明,TSA平皿在暴露一段时间后,由于水分的流失影响微生物的生长,培养基灌装量大的TSA平皿耐受的暴露时间较长。 To envaluate the microbial growth of TSA culture, which is exposed for a short time in clean environmental of pharmaceutical enterprises, comparative experiments were carried out using two kinds of specifications, three manufacturers, two time method.The results were that after exposure to 2.1h, loading capacity of 25 m L and 30 m L TSA was suitable for Staphylococcus aureus, Pseudomonas aeruginosa, Bacillus subtilis, Candida, Aspergillus niger growth,but after exposure to 4.1h, loading capacity of 25 m L TSA was not suitable for Microorganism growth. The results showed that the TSA plate was exposed for a period of time, due to the loss of water affected the growth of microorganisms, the exposure time of the TSA plate with large filling capacity was longer.
