摘要
采用SYKAM S501 Series液相色谱仪,Shimadzu Shim-pack VP-ODS C18色谱柱(5μm,4.6mm×250mm),甲醇∶水=35∶65为流动相,检测波长为250nm,快速测定葛根酒中葛根素含量,并通过超氧阴离子自由基(O_2^-·)、羟基自由基(·OH)、1,1-二苯基-2-苯基自由基(DPPH·)清除率测定法评价葛根酒体外抗氧化活性。结果表明,葛根酒中葛根素的含量为160.83μg/mL。其对1,1-二苯基-2-苯基自由基的清除率达到53.1%,对羟基自由基的清除率达到35.67%,对超氧阴离子自由基的清除率达到75.36%。这说明葛根酒中含有较多的葛根素,具有较强的体外抗氧化活性,并具有一定的保健作用。
SYKAM S501 Series High Performance Liquid Chromatography was used for rapid measurement of puerarin content in pueraria wine(Shimadzu Shim-pack(5 μm,4.6 mm× 250 mm), methanol ∶ water =35 ∶ 65 as mobile phase, and detection wavelength was 250 nm), and through measuring scavenging rate of ultra oxygen anion free radical(O_2^-·), hydroxyl radicals(·OH) and 1, 1-diphenyl-2-phenyl-free radical(DPPH·) was to evaluate the antioxidant activity of pueraria wine in vitro. The results showed that the content of puerarin in pueraria wine was 160.83μg/mL and the scavenging rate of DPPH·, ·OH and O_2^-·free radical reached 53.1%, 35.67% and 75.36% respectively. This shows that puerarin wine contains many puerarin compound, which has strong antioxidant activity in vitro and has healthcare functions.
