摘要
从陈年谷酒酒醪中提取一株高产纤维素酶的菌株,利用形态和16SrDNA基因序列进行分析,确定该菌株为甲醇芽胞杆菌(Bacillus methylotrophicus)。利用产酶发酵培养基培养产生纤维素酶,研究碳源、氮源、培养基初始pH、起始温度、培养时间和接种量对菌株产酶的影响。结果表明,菌株最合适的发酵条件为:最佳碳源为CMC-Na,最适氮源为(NH_4)_2SO_4,最适pH为5.0,最适发酵时间为5d,最适温度为30℃。从而筛选出高产纤维素霉菌株,优化了发酵条件。
A high yield cellulase strains was extracted from the old Valley wine mash filter, morphological and 16 SrDNA gene sequences were used for analysis, the strain was identified as Bacillus methylotrophicus. Cellulase production was carried out by fermentation medium, and the effects of carbon source, nitrogen source, initial pH, initial temperature, incubation time and inoculation amount on the enzyme production were studied. The results showed that the optimal fermentation conditions were as follows: the optimum carbon source was CMC-Na, the optimum nitrogen source was(NH_4)_2SO_4, the optimum pH was 5, the optimum fermentation time was five days, and the optimum temperature was 30 ℃.
