离子交换法制备硫酸卷曲霉素工艺

摘要 卷曲霉素(Capreomycin)是一种碱性水溶性抗生素,临床上常用其硫酸盐,即硫酸卷曲霉素,离子交换法是提取卷曲霉素的常用工艺之一。本文对用弱酸性离子交换树脂提取硫酸卷曲霉素工艺进行研究。考察了防腐剂(甲醛)与卷曲霉素的相互作用,从离子交换体系的酸碱度、树脂饱和度以及其他相关因素入手,研究各工艺参数或相关因素对产品质量的影响。结果显示:甲醛对卷曲霉素主要成分有破坏作用,可使卷曲霉素CA1组分下降,进而影响产品含量的提高;将系统pH提升至9.0左右,不仅可以延缓因系统杂菌滋生导致pH下降而且更利于卷曲霉素含量的提高;利用废液组分分析法判断树脂饱和度更利于质量控制与稳定。 Capreomycin is an alkaline water-soluble antibiotic. Commonly its sulfate is used in clinic. Ion exchange is one of the common processes for extracting capreomycin. In this paper, the ion exchange technology of weakly acidic resin was studied. Effects of preservatives(formaldehyde) interaction with capreomycin is investigated. From the ion exchange system p H, resin saturation and other related factors, studied the influence of various process parameters or related factors on the quality of the products. The results showed that, formaldehyde has a destructive effect on the main components of Capreomycin, which can reduce the component of CA1, thereby debase the content of the product. It is not only delay the decline of p H by the multiplication of bacteria in the system, but also benefit the content of capreomycin by up the system p H to 9. It is better for quality control and stability to judge resin saturation by using component analysis of waste liquid.
