一种适用于HPLC去除血清中干扰峰的方法

摘要 通过比较和优化血清样品制备方法,建立一种适用于HPLC去除血清中干扰峰的方法。运用盐析法、加热法、萃取法和有机溶剂法去除血清中干扰峰,经HPLC分析,比较4种方法对血清中干扰峰的去除效果。结果表明乙腈沉淀法能去除血清中大部分的蛋白质,留下很少的杂质峰;而加热法、萃取法和有机溶剂法均能去除血清中的杂质峰,但是留下的干扰峰较多,不适用于血清中HPLC的其他物质的检测。乙腈沉淀法去除血清中的杂质峰效果最好,干扰最小,为HPLC血清中杂质峰的排除提供了一种优化的方法。 Objectives: By comparing and optimizing the preparation method of serum samples, a method for removing the interference peak in serum by HPLC was established. Methods: The interference peak was removed by salting out method, heating method, extraction method and organic solvent method. The removal efficiency of serum interference peak was analyzed by HPLC. Results: The acetonitrile precipitation method could remove most of the protein in the serum, leaving little impurity peak. The heating method, the extraction method and the organic solvent method could remove the impurity peaks in the serum, but the interference peak was more, does not apply to the detection of other substances in the serum HPLC.Conclusion:The acetonitrile precipitation method to remove impurities in the peak of the best results, the smallest interference. Providing an optimized method for the further elimination of impurity peaks inserumby HPLC.
