摘要
真核生物基因通过可变转录起始位点、可变剪切和可变多聚腺苷酸化产生多个转录本异构体(transcript isoform)。由于不同转录本异构体的翻译情况有时会有较大的不同,这就导致转录本的多样性和蛋白产物之间的对应关系比较复杂。本文中,笔者收集了果蝇早期胚胎细胞转录组数据和翻译组数据,并通过系统的分析展示了不同细胞组分转录本组成异同。通过对转录组数据和翻译组数据进行比较分析,一共发现766个基因的转录本相对表达水平在果蝇不同细胞组分中发生了改变,这表明在果蝇中大量基因的不同转录本的翻译效率差异可能很大。
By using alternative transcription start site,alternative splicing and alternative polyadenylation,eukaryotic genes produce multiple transcript isoforms.The translation efficiency of each transcript is sometimes different,leading to complex relationship between transcript diversity and protein products.We systematically compared the translatome sequencing data of the early embryo of Drosophila with the corresponding transcriptome sequencing data,revealing the difference between the transcript composition of different cellular compartments.Through the analysis of the transcriptome and translatome data,we found 766 genes whose relative transcript isoform abundance changed between different cellular compartments in Drosophila,indicating that different transcripts of many genes in Drosophila may have very different translation efficiency.
