摘要
目的:观察Pokemon基因敲减后Lovo细胞对药物(dehydrocostus lactone,DHC)增殖抑制敏感性的变化。方法:构建Pokemon敲减的Lovo细胞稳定细胞株Lovo-KD,用荧光定量PCR检测Pokemon基因敲减效率,分别在Lovo-NC、Lovo-KD细胞中加入各浓度梯度的去氢木香内酯(dehydrocostus lactone,DHC),分别在24h、48h、72h用MTT法检测细胞增殖抑制效率。结果:与Lovo-NC组细胞相比,相同梯度DHC(10、15、20μg/ml)对Lovo-KD细胞24h、48h、72h的增殖抑制率均显著升高,如20μg/ml DHC作用48 h后Lovo-KD、Lovo-NC组细胞增殖抑制率分别为82.11±2.3%、28.93±2.4%,P<0.01。结论:Pokemon基因敲减可增加Lovo细胞对DHC药物增殖抑制敏感性。
Objective: to observe the change of proliferation inhibition sensitivity to DHC drugs in Pokemon knockdown Lovo cells. Methods: Stable Pokemon knockdown Lovo cell line(LOVO-KD) and the negative control Lovo cell line(Lovo-NC) were constructed by transfecton of the shRNA plasmids, respectively. The mRNA level of Pokemon was measured with real-time PCR. Detect the change of inhibition action on proliferation activity by MTT assay. Results: compared with Lovo-NC group, treated with same concentration gradient DHC, the proliferation inhibition rate of Lovo-KD was increased significantly at the time point of 24, 48,and 72 hours. Lovo-KD proliferation inhibiton rate treated with 20μg/ml DHC for 48 hours was 82.11±2.3%, while the Lovo-NC was 28.93±2.4%, P<0.01. Conclusion: Pokemon knockdown in Lovo cells could enhanced the proliferation inhibition sensitivity of DHC drug.
