几种水果对小鼠肝脏和肠道Cyp3a11代谢活性的影响

摘要 目的:研究几种水果对小鼠Cyp3a活性的影响,避免药物相互作用。方法:将供试物给小鼠灌胃2次/d,连续7d。用超速离心法分离小鼠肝/肠微粒体;分光光度法检测微粒体中Cyp3a11活性。结果:以红霉素和氨基吡啉为底物测定肝脏和肠道Cyp3a活性时,罗汉果和广东柚子高、低剂量组小鼠Cyp3a活性明显高于纯水对照组(P<0.01或P<0.05);而龙眼、杨桃和木瓜各剂量组对小鼠Cyp3a活性没有影响。结论:罗汉果和广东柚子对小鼠肝脏和肠道Cyp3a11的活性均有诱导作用。本实验结果提示,罗汉果和广东柚子有可能增强经CYP3A4代谢的各种药物在体内的代谢。 Objective: Aim to investigate the effect of several tropical fruits on the Cyp3a11 activity in mouse so that support a direction for drug-drug interaction. Method: The mice were randomly divided into thirteen group, the mice of each group were treated with corresponding fruit juice two times a day for seven days. The concentration of protein in microsome was determined by the Bradford method. The activity of Cyp3a11 was detected by spectrophotometer.Result: the fruits effect on Cyp3a11 activity in liver and intestine,erythromycin and aminopyrine as the substrate, Momordica grosvenori and Pomelo groups have significant difference with control group(P<0.05, P<0.01); the other fruits have no effects on Cyp3a11 activity in mice. Conclusion: Momordica grosvenori and Pomelo can induce the activity of Cyp3a11 in mice liver and intestine.
