摘要
目的:研究肺结核患者血浆纤维胶凝蛋白-2(Ficolin-2)的表达及与T淋巴细胞亚群、NK细胞间的关系。方法:研究对象为101例肺结核患者和116例健康对照者,采用酶联免疫吸附实验(ELISA)检测血浆Ficolin-2水平,采用流式细胞术检测CD3+、CD4+、CD8+T淋巴细胞以及NK细胞的百分数。结果:肺结核患者血浆Ficolin-2水平明显低于健康对照组(4.95±1.30vs.10.36±4.46,P<0.01),CD3+T淋巴细胞(%)、CD4+T淋巴细胞(%)和CD8+T淋巴细胞(%)与健康对照组相比也显著降低(均P<0.01),但NK细胞(%)与对照组相比差异无统计学意义;进一步研究发现,肺结核患者血浆Ficolin-2水平与CD3+T淋巴细胞、CD4+T淋巴细胞呈正相关(Pearson Correlation=0.549,P<0.01和Pearson Correlation=0.551,P<0.01),与CD8+T细胞、NK细胞未见明显相关性。结论:肺结核患者血浆Ficolin-2下降伴随着细胞免疫功能降低,推测Ficolin-2的降低可能加速结核分枝杆菌感染者的疾病进程。
Objective:To investigate the changes of plasma ficolin-2 and T lymphocyte subsets in tuberculosis patients,analyze the relationships between them and discuss the significance.Method:Ficolin-2 levels and T lymphocyte subsets of 116 tuberculosis patients and 101 healthy controls were measured by enzyme-linked immunosorbent assay(ELISA)and flow cytometry(FCM),respectively.Result:Plasma ficolin-2 level of tuberculosis patients was lower than that of healthy controls(4.95±1.30 vs.10.36±4.46,P<0.01).The percentages of CD3+,CD4+and CD8+T cells as well as NK cells decreased significantly in tuberculosis patients than that in controls(all P<0.01).Ficolin-2 level was positively related to the percentages of CD3+and CD4+T cells(Pearson Correlation=0.549,P<0.01 and Pearson Correlation=0.551,P<0.01,repectively),and was not related to the concentrations of CD8+T cells and NK cells.Conclusion:Low expressions of ficolin-2 might be associated with decreased cellular immunity in tuberculosis patients,which might play an important role in the early infection and development of disease.
