摘要
[目的]研究不同激素种类和浓度配比对晋谷21号谷子种子愈伤组织诱导及植株再生的影响,旨在筛选最佳的愈伤组织诱导和分化的激素配比,建立稳定高效的谷子种子再生体系。[方法]以晋谷21号谷子的成熟种子为外植体,研究不同激素种类和浓度配比对晋谷21号谷子愈伤组织诱导及植株再生的影响。[结果]愈伤组织的诱导以MS培养基中附加2,4-D的效果要优于NAA,同时加入适合浓度的6-BA与之配比可以提高愈伤组织的质量,以MS培养基中附加2,4-D 2.0 mg·L^(-1)、6-BA 0.2 mg·L^(-1)时,愈伤组织诱导率最高,达92.86%; 6-BA诱导愈伤组织分化的效果优于ZT,培养基中附加6-BA 1.0 mg·L^(-1)时,愈伤分化率最高,为13.33%;将谷子分化苗转接于MS培养基上可诱导生根。[结论]诱导愈伤组织的最佳培养基为MS+2.0 mg·L^(-1)2,4-D+0.2 mg·L^(-1)6-BA,愈伤组织分化的最佳培养基为MS+1.0 mg·L^(-1) 6-BA。
[Objective] Effects of varioustypes of hormones at different concentrations on callus induction and plant regeneration of Jingu 21 seedlings were investigated. Present study was aimed to determine the optimal hormone ratio for better callus induction and differentiation, and to establish a stable millet regeneration system with high efficiency. [Methods] The matured seeds of Jingu 21 millet were employed as explant material, and effects of different types of hormones at different concentrations on callus induction and plant regeneration of Jingu 21 were studied. [Results] By adding 2,4-D to MS medium, the callus induction rate was enhanced and better than that of NAA, and the quality of callus was improved by adding a certain concentration of 6-BA to the medium.When the concentration of 4-D and 6-BA were 2.0 mg·L-1 and 0.2 mg·L-1, respectively, the callus induction rate reached the highest peak at 92.86%. In the process of callus differentiation, the effect of adding 6-BA was significantly better than that of ZT, with the highest callus differentiation rate(13.33%) occurred at the concentration of 6-BA at 1-0 mg·L-1. In addition, transferring millet seedlings to MS medium induced root development. [Conclusion] The optimal medium for inducing callus was MS mixed with 2.0 mg·L-1 2,4-D and 0.2 mg·L-16-BA, and the best differentiation medium combination was MS plus 1.0 mg·L-1 6-BA.
