摘要
目的探讨葛根素对2型糖尿病小鼠心梗后心肌能量代谢的影响。方法建立糖尿病併心梗小鼠模型,将动物分为糖尿病假心梗组(shame);糖尿病心梗组(MI);葛根素组(MI+PUE),第3组小鼠腹腔注射葛根素1. 3mL·kg^(-1)·d^(-1),治疗4周,统计3组小鼠的生存率。提取各组小鼠心肌组织总蛋白和膜蛋白,取各组蛋白样品20μg,用Western blot方法检测心肌细胞GLUT4和CD36蛋白的表达。结果结扎后的四周,MI组超过一半的小鼠死亡。MI+PUE组生存率较MI组显著提高(P=0. 048);糖尿病併冠脉结扎小鼠,总GLUT4和m GLUT4蛋白均增多;总CD36以及m CD36蛋白均减少;与MI组比较,MI+PUE组的GLUT4和m GLUT4增多而CD36和m CD36减少。结论葛根素通过调控能量代谢来改善糖尿病併冠脉结扎小鼠的心脏功能,其潜在机制可能是葛根素提高了GLUT4的转运和表达,同时减弱了CD36的转运和表达。
Objective To investigate the effect of puerarin on myocardial energy metabolism in type 2 diabetic mice after myocardial infarction. Method The mice model of diabetic myocardial infarction were established and divided into diabetic pseudomyocardial infarction group( shame),diabetic myocardial infarction group( MI),puerarin group( MI +PUE). The mice in group 3 were injected puerarin 1. 3 m L·kg-1·d-1 intraperitoneally for 4 weeks,and the survival rates of the three groups were counted. Total and membrane proteins were extracted from myocardial tissues of mice in each group. The expression of GLUT4 and CD36 proteins in myocardial cells was detected by Western blot. Result Four weeks after ligation,more than half of the MI mice died. The survival rate of MI + PUE group was significantly higher than that of MI group( P = 0. 048);the total GLUT4 and m GLUT4 protein increased in diabetic mice with coronary ligation;the total CD36 and m CD36 protein decreased;compared with MI group,the GLUT4 and m GLUT4 increased in MI+ PUE group,while the CD36 and m GLUT36 decreased. Conclusion Puerarin improves cardiac function in diabetic mice with coronary artery ligation by regulating energy metabolism. Potential mechanism is that puerarin increases the transport and expression of GLUT4 and decreases the transport and expression of CD36.
