摘要
目的:探讨非小细胞肺癌组织中ABCE1与MMP-9表达及其二者之间的相关性。方法:采用免疫组化和qRT-PCR技术检测30例非小细胞肺癌组织及对应的癌旁组织中ABCE1、MMP-9的表达水平,分析其在非小细胞肺癌组织中的表达及相关性。结果:癌组织中ABCE1和MMP-9蛋白的表达明显高于癌旁组织。ABCE1和MMP-9 mRNA在癌组织中的表达也明显高于癌旁组织(P<0.001)。高表达ABCE1 mRNA癌组织中MMP-9 mRNA表达也较高,二者之间存在正相关(r=0.596,P=0.001)。结论:在非小细胞肺癌组织中,ABCE1基因可能通过提高MMP-9的表达从而促进了NSCLC的侵袭转移过程。
Objective: To detect the expression correlation of ATP- binding cassette transporter E1( ABCE1) and matrix metallopeptidase- 9( MMP- 9) in human non- small cell lung cancer. Methods: The immunohistochemistry and realtime RT- PCR were used to detect the expression of mRNA and protein of ABCE1 and MMP- 9 in lung cancer and corresponding adjacent tissues of 30 non- small cell lung cancer patients. Results: The expression of ABCE1 and MMP- 9 protein were significantly higher in the cancer tissue than corresponding adjacent tissues. The expression of ABCE1 and MMP- 9 mRNA were significantly higher in the cancer tissue than corresponding adjacent tissues( P < 0. 001). ABCE1 was positively correlated with MMP- 9 in lung cancer tissues( r = 0. 596,P = 0. 001).Conclusion: ABCE1 gene may accelerate the process of invasion and metastasis of NSCLC via increasing the expression of MMP- 9 in non- small cell lung cancer.
