摘要
中心体是哺乳动物细胞内的微管组织中心,其主要功能为参与纺锤体的装配,在胞质分裂中起重要调控作用。最新发现的中心体相关蛋白CEP55(centrosomal protein,55 kD)属于卷曲螺旋(coiled-coil)蛋白质家族成员,其主要功能是锚定微管聚合相关蛋白,参与纺锤体形成,进而调控细胞增殖。该蛋白质在正常组织及肿瘤细胞中均存有表达,与细胞周期中的中心体和中间体相偶联,磷酸化后发挥调控细胞周期的作用。已经发现CEP55过表达与很多恶性肿瘤的肿瘤分期、侵袭性、肿瘤转移等有显著相关性。本文中我们通过讨论CEP55在不同效应途径中的功能及其调控因子等,明确CEP55作为肿瘤发生、进展、预后的生物学标志物的作用。
The midbody is the microtubule organizing center in mammalian cells. Its main function is involved in spindle assembly and it is a key regulator of cytokinesis. CEP55( centrosomal protein,55 kD) is a latest found mitotic phosphoprotein,and CEP55 is a member of the coiled-coil protein family. Its main function is anchoring the microtubule polymerization-related proteins,involving in spindle formation,then regulaing cell proliferation. The proteins are expressed both in normal tissues and tumor cells. CEP55 is coupled with the centrosome and midbody during the cell cycle. It play an important role in cell cycle regulation after phosphorylation. CEP55 overexpression has been found to significantly correlate with tumor stage,aggressiveness,metastasis and poor prognosis in multiple tumor types. In this study,we examined the role of CEP55 as a biomarker of tumorigenesis,progression and prognosis by discussing the role of CEP55 in different pathways and its regulatory factors.
