摘要
目的探讨DJ-1基因在肺癌发生发展中的作用。方法选取肺鳞癌细胞株SK-MES-1和HTB-182细胞株。构建DJ-1siRNA,siRNA感染细胞。采用WB验证DJ-1siRNA的干扰情况,采用MTT检测各组细胞的增殖能力。观察DJ-1 si RNA干扰情况、各组细胞的增殖能力。结果 DJ-1 siRNA干扰组中DJ-1的蛋白质表达水平显著低于NC组和BC组,DJ-1 siRNA组细胞增殖速度显著慢于NC组和BC组,差异具有统计学意义(P<0.05)。结论 DJ-1基因能够抑制肺鳞癌细胞增殖,阻碍相关蛋白的表达。
Objective To explore the role of DJ-1 gene in the development of lung cancer. Methods SK-MES-1 and HTB-182 were selected. DJ-1siRNA and siRNA infection cells were structured. The interference of DJ-1siRNA were tested by WB,the cell proliferation were tested by MTT. The DJ-1siRNA interference and the cell proliferation were observed. Results The protein expression of DJ-1 in DJ-1siRNA group was lower than those of NC group and BC group( P < 0. 05). The cell proliferation rate of DJ-1siRNA group was lower than those of NC group and BC group( P < 0. 05). Conclusion The DJ-1 can inhibit lung squamous cancer cell proliferation,and hinder the related protein expression.
