大肠癌淋巴结转移与PTEN、COX-2表达的相关性 被引量:7

摘要 目的探讨大肠癌淋巴结转移与PTEN、COX-2表达的相关性。方法选取手术切除所得100例患者的大肠癌组织和正常大肠组织,采用免疫组化法检测大肠癌组织和正常大肠组织中PTEN、COX-2的表达。统计分析组织来源者淋巴结转移情况并分析大肠癌淋巴结转移与PTEN、COX-2表达的相关性。结果大肠癌组织中PTEN的阳性表达率为52%,显著低于正常大肠组织的100%;而大肠癌组织中COX-2的阳性表达率为92%,显著高于正常大肠组织的6%,差异有统计学意义(P<0.01)。PTEN阳性表达患者淋巴结转移发生率明显低于PTEN阴性表达患者,而COX-2阳性表达患者的淋巴结转移发生率则明显高于COX-2阴性表达患者,差异有统计学意义(P<0.01)。Spearman关系分析结果显示,大肠癌淋巴结转移与其PTEN的阳性表达率呈负相关(γ=-0.758,P<0.05),与其COX-2阳性表达率则呈正相关(γ=0.792,P<0.05)。结论大肠癌淋巴结转移与其PTEN的阳性表达率呈负相关,与其COX-2阳性表达率则呈正相关,PTEN的阴性表达和COX-2的阳性表达可预测大肠癌患者淋巴结转移的发生。 Objective To explore the relationship between lymph node metastasis of colorectal cancer and expression of PTEN and COX-2. Methods Colorectal cancer tissues and normal colon tissues from 100 colorectal cancer patients were selected. Immunohistochemical method was used to detect the expression level of PTEN and COX-2 in colorectal cancer tissues and normal colon tissues. Lymph node metastasis of the patients was statistical analyzed and the relationship between PTEN and COX-2 expression and lymph node metastasis of colorectal cancer patients were analyzed. Results The positive expression rate of PTEN in colorectal cancer tissues was 52%,which was significantly less than 100% of the normal colon tissues,and COX-2 positive expression rate of colorectal cancer tissues was 92%,which was significantly higher than 6% of the normal colon tissues,the difference was statistically significant( P < 0. 01). Lymph node metastasis rate of PTEN positively expressed patients was obviously lower the PTEN negatively expressed patients,while lymph node metastasis rate of patients with COX-2 positive expression was significantly higher than that of COX-2 negatively expressed patients,the difference was statistically significant( P < 0. 01).Spearman relationship analysis showed that colorectal cancer lymph node metastasis and positive expression rate of PTEN was negatively correlated( γ =- 0. 758,P < 0. 05),and it was positively correlated with patients with COX-2 positive expression( γ =0. 792,P < 0. 05). Conclusion Lymph node metastasis of colorectal cancer is negatively correlated with positive expression rate of PTEN,and positively correlated with COX-2 positive expression rate. Negative expression of PTEN and positive expression of COX-2 can predict the occurrence of lymph node metastasis in patients with colorectal cancer.
