摘要
针对实时荧光定量PCR仪光学校准方法的不完善,通过对实时荧光定量PCR仪校准必要性、基本结构和数学原理的分析,基于熔解曲线从C_t值均匀度、C_t值精密度、通道峰值高度一致性、线性灵敏系数、熔解温度漂移和熔解温度比等方面对实时荧光定量PCR仪校准方法进行分析。
In view of the imperfect of optical calibration method of real-time fluorescent quantitative PCR analyzer,by analyzing the necessity of calibration,basic structure and mathematical principle of real-time fluorescent quantitative PCR analyzer,the calibration method of real-time fluorescent quantitative PCR analyzer was analyzed from the aspects of C_tvalue uniformity,C_tvalue precision,channel peak height consistency,linear sensitivity factor,melt temperature bias,and ratio of melt temperature based on the melt curve.
