摘要
目的建立SD大鼠胎盘提取及胎盘细胞培养的实验方法,为胎盘细胞的研究奠定基础。方法颈椎离断处死孕期20 d的SD大鼠,提取大鼠胎盘,选取有活性的胎盘组织进行细胞培养,采用倒置显微镜观察细胞形态。结果解剖孕期20 d的SD大鼠可取得胎盘,细胞培养后胎盘细胞贴壁生长,细胞形态为多角形或梭形,边界清楚,细胞核为圆形或椭圆形。结论本实验建立的实验方法切实可行,为胎盘细胞研究提供了具体的实验方法,为胎盘细胞的研究奠定基础。
Objective To establish experimental method for SD rat placenta extract and cell culture so as to lay foundation for the placental cell research. Method Push 20 days of gestation SD rats to death to extract rat placentas and culture cells were isolated from active placental tissue,adopt inverted microscope to observe cell morphology. Result Placentas can be obtained by dissecting 20 days of gestation SD rats,placental cells adhered under the cells culture. Cell morphology was polygonal or fusiform,cells had defined margins and abundant cytoplasm. Nucleus was circular and elliptical. Conclusion Experimental method is feasible in this experiment.This experiment provide specific experimental method for the research of placental cells,and lay foundation for the placental cell research.
