摘要
目的建立利用DNA稳定的银纳米簇检测端粒酶活性的方法。方法利用合成的寡核苷酸序列能稳定银纳米的特点,在强还原剂硼氢化钠存在下还原银离子得到具有良好荧光特性的银纳米簇。结果利用端粒酶活性能够延伸5'-(TTAGGG)n-3'的特性,以及含有G碱基的DNA序列靠近银纳米簇合成模板时,合成的银纳米簇的荧光明显提升特性建立端粒酶活性检测的方法。结论合成了以寡核苷酸为模板的银纳米簇并对其荧光特性进行表征,建立了DNA稳定的银纳米簇用于检测端粒酶活性的方法。
Objective To establish a method to detect telomerase activity using DNA-templated fluorescent silver nanoclusters. Method In the presence of sodium borohydride and synthesized oligonucleotide,silver ions in the nanoparticles were reduced into silver nanoclusters with excellent fluorescent properties. When DNA sequences containing G bases connected with the DNA-templated silver nanoclusters,the fluorescence will be improved obviously. Telomerase activity was detected using the oligonucleotide 5'-( TTAGGG)n-3',which could be extended by telomerase. Result and Conclusion The method for telomerase activity detection was established using the DNA-templated fluorescent silver nanoclusters.
