摘要
研究了胡椒中胡椒碱的含量采用高效液相色谱法检测。胡椒试样研磨成粉状,过筛经乙酸-乙醇(15:85)混合溶液超声提取后,以甲醇-水为流动相,采用等度洗脱方式,二极管阵列检测器进行检测。胡椒碱在40~400.0 mg/L线性范围内呈现良好的线性关系,相关系数为0.999 7。在添加一定浓度水平条件下,胡椒碱的平均回收率为73.2%~81.9%,相对标准偏差为6.13%~7.08%,方法检出限为1.35 mg/L,方法定量限为4.67 mg/L。经方法学验证,混合溶剂超声提取法具有操作简单、检测成本低,能在短时间内实现大批量操作,为相关的检测方法研究开发提供了参考依据。
In this study,high performance liquid chromatography(HPLC)was used to determine piperine in pepper.Samples were sieved into powder and screened by ultrasonic extraction of acetic acid and ethanol(15:85)mixture.The methanol and water were used as mobile phase,and piperine was separated by isocratic elution and finally detected with diode array detector.The linear relationship was observed in the linear range of 40~400.0 mg/L,and the correlation coefficient(r)was 0.999 7.The average recoveries of piperine were 73.2%~81.9%and the relative standard deviations(RSD)were 6.13%~7.08%under certain conditions.The detection limit was 1.35 mg/L and the limit of quantification was 4.67 mg/L.The method has proved that the mixed solvent ultrasonic extraction method has simple operation,low detection cost,and can realize large-scale operation in a short time,which provides reference for the research and development of related detection methods.
