摘要
以2-氯烟酸、4-哌啶甲酸甲酯为原料反应合成2-[N-(4-甲氧基甲酰基)哌啶基]-3-吡啶甲酸乙酯,并对合成的产物通过TLC跟踪、测定熔点、红外光谱以及核磁氢谱迚行结构表征,通过实验讨论了4-哌啶甲酸甲酯与中间体2-氯烟酸乙酯的配料比、反应温度对产物收率的影响,确定最佳配料比2-氯烟酸乙酯∶4-哌啶甲酸甲酯=1∶1.2,反应温度为110℃,同时确定最佳合成路线。
Ethyl 2-[N-(4-methoxyformyl)piperidinyl]-3-pyridinecarboxylate was synthesized by the reaction of 2-chloronicotinic acid and methyl 4-piperidinecarboxylate.The structure of synthesized product was characterized by TLC tracking,determination of melting point,IR and 1 H-NMR.The effect of the formulation ratio of methyl 4-piperidinecarboxylate to the intermediate 2-chloronicotinic acid ethyl ester,and the reaction temperature on the product yield was investigated through experiments.The best conditions were determined as follows:ethyl 2-chloronicotinate:methyl 4-piperidinecarboxylate=1∶1.2,the reaction temperature 110℃,meanwhile the best synthetic route was also determined.
