摘要
对一株采自云南省的野生真菌进行菌丝分离,采用形态学与ITS法对其进行鉴定,鉴定结果为隐孔菌(Cryptoporus volvatus)。采用体外间接酶连免疫荧光法对隐孔菌菌丝提取物进行抗病毒活性检测,结果表明,菌丝提取物具有抗猪繁殖和呼吸障碍综合症(porcine reproductive and respiratory syndrome,PRRS)病毒的作用。采用单因素实验对隐孔菌最适液体培养条件和碳氮源进行研究,结果表明,最适培养条件为26℃,pH 4~7,装液量为50 mL(250 mL三角瓶);最适碳源和氮源分别为葡萄糖和黄豆饼粉。采用正交实验对隐孔菌最佳营养元素配比进行研究,结果表明,最佳组合为80 g/L葡萄糖、45 g/L黄豆饼粉、1 g/L K_2HPO_4·3H_2O、0.25 g/L MgSO_4·7H_2O。
A wild fungal strain was collected,isolated, cultured and identified by morphological and Internal Transcribed Spacer(ITS) methods as Cryptoporus volvatus in this study. The extracts from mycelium were proved to have antiviral activities against porcine reproductive and respiratory disorders virus by an indirect enzyme-linked immunofluorescence assay in vitro. Univariate experiment was used to study the optimal liquid culture conditions, carbon and nitrogen sources of C. volvatus. Results showed that the optimal culture conditions were 26 ℃,pH 4.0~7.0 and 50/250 mL liquid loading,and the optimal carbon and nitrogen sources were glucose and soybean cake powder, respectively. Orthogonal experiment was applied to test the optimum ratio of nutrients. The optimal formula was: 80 g/L glucose, 45 g/L soybean cake powder, 1 g/L K2 HPO4·3 H20 and 0.25 g/L MgSO4·7 H2 O.
